Objectives of this study were twofold: (i) to assess the association between polymorphonuclear (PMN) counts and chronic alterations within the bovine endometrium and (ii) to determine the distribution of inflammation throughout the endometrium of clinically healthy dairy cows. Holstein‐Friesian cows (n = 32) from a single dairy farm were selected for this experiment. Before slaughtering, a complete reproductive examination was performed to discard any type of clinical disease. After slaughtering, reproductive tracts were collected, and the endometrium was sampled at 8 pre‐defined locations. At each location, endometrial biopsies (EBs) and cytology (CY) samples were harvested. Histopathology samples were stained with haematoxylin–eosin (EB‐HE) and naphthol‐AS‐D‐chloroacetate‐esterase (EB‐naphthol), while CY samples were stained with Wright–Giemsa. In the EB‐HE samples, parameters assessed were epithelium height, mononuclear cells infiltration, lymphocytic aggregates, periglandular fibrosis, angiosclerosis and haemorrhage. In EB‐naphthol and CY slides, PMNs counts were evaluated. Binomial logistic regression was used to assess the association between the number of PMNs present in both the EB‐naphthol and CY samples and alterations identified in the EB‐HE samples and to analyse the distribution of the histopathological alterations (EB‐HE). A Poisson mixed‐effect model was used to analyse the distribution of PMNs within the endometrium. A significant positive association was found between the PMN counts and the mononuclear cells infiltration. The presence of erythrocytes was associated with higher odds to detect PMNs in the stratum compactum. Significantly, higher infiltration of PMNs and mononuclear cells were detected in the uterine body and the right horn region. Concluding, CY is a technique that allows the evaluation of PMN counts and therefore only evaluates active inflammation. A complete assessment of endometrial health can only be obtained using EB. To optimize the sensitivity to diagnose endometrial inflammation in cows, adjacencies of the corpus uteri should be considered as the preferred region to harvest samples.  相似文献

BACKGROUND: Because bedbugs, Cimex lectularius L., have not been a problem in the USA for over 40 years, few insecticide products are labeled for their control. Most products that are labeled for bedbugs are pyrethroids. However, recent studies indicate that field‐collected bedbugs may be resistant to pyrethroids. There are also non‐pyrethroid products labeled for bedbugs, but, like the pyrethroids, none of these products has been evaluated for field efficacy. This study evaluated the efficacy of two insecticide treatment regimens for bedbugs in multi‐unit housing. Both of the treatments included multiple products currently being used by the pest management industry. RESULTS: The ‘traditional’ treatment consisted of applications of pyrethroid products and an insect growth regulator. The ‘novel’ treatment consisted of applications of non‐pyrethroid products. The traditional treatment significantly reduced the number of bedbugs from 39.8 ± 10.1 per unit prior to treatment to 2.2 ± 1.0 at the end of the test period (P = 0.02; 95% reduction). The number of live bedbugs in the ‘novel’ treatment was also significantly reduced from 71.4 ± 25.3 bedbugs per unit to 10.2 ± 4.4 after 8 weeks (86% population reduction). CONCLUSIONS: Although both treatment regimens reduced bedbug numbers, the fact that bedbugs were not eliminated after multiple applications suggests that the insecticides, applied at the current label rates, were inadequate. Mangrove ecosystems are tropical environments that are characterized by the interaction between the land and the sea. As such, this ecosystem is vulnerable to oil spills. Here, we show a culture-independent survey of fungal communities that are found in the sediments of the following two mangroves that are located on the coast of Sao Paulo State (Brazil): (1) an oil-spill-affected mangrove and (2) a nearby unaffected mangrove. Samples were collected from each mangrove forest at three distinct locations (transect from sea to land), and the samples were analyzed by quantitative PCR and internal transcribed spacer (ITS)-based PCR-DGGE analysis. The abundance of fungi was found to be higher in the oil-affected mangrove. Visual observation and correspondence analysis (CA) of the ITS-based PCR-DGGE profiles revealed differences in the fungal communities between the sampled areas. Remarkably, the oil-spilled area was quite distinct from the unaffected sampling areas. On the basis of the ITS sequences, fungi that are associated with the Basidiomycota and Ascomycota taxa were most common and belonged primarily to the genera Epicoccum, Nigrospora, and Cladosporium. Moreover, the Nigrospora fungal species were shown to be sensitive to oil, whereas a group that was described as ??uncultured Basidiomycota?? was found more frequently in oil-contaminated areas. Our results showed an increase in fungal abundance in the oil-polluted mangrove regions, and these data indicated potential fungal candidates for remediation of the oil-affected mangroves.  相似文献

Mangroves are intertidal extreme environments with rich microbial communities. Actinobacteria are well known for producing antibiotics. The search for biosynthetic potential of Actinobacteria from mangrove environments could provide more possibilities for useful secondary metabolites. In this study, whole genome sequencing and MS/MS analysis were used to explore the secondary metabolite production potential of one actinobacterial strain of Streptomyces olivaceus sp., isolated from a mangrove in Macau, China. The results showed that a total of 105 gene clusters were found in the genome of S. olivaceus sp., and 53 known secondary metabolites, including bioactive compounds, peptides, and other products, were predicted by genome mining. There were 28 secondary metabolites classified as antibiotics, which were not previously known from S. olivaceus. ISP medium 2 was then used to ferment the S. olivaceus sp. to determine which predicted secondary metabolite could be truly produced. The chemical analysis revealed that ectoine, melanin, and the antibiotic of validamycin A could be observed in the fermentation broth. This was the first observation that these three compounds can be produced by a strain of S. olivaceus. Therefore, it can be concluded that Actinobacteria isolated from the mangrove environment have unknown potential to produce bioactive secondary metabolites.  相似文献

The strong hunting pressure on the red-legged partridge, Alectoris rufa, warranted its inclusion into the list of species of European conservation concern. During the last decades, restocking plans with farmed specimens have counterbalanced the hunting drawings from wild populations. Our concern was the study of A. rufa in the easternmost part of its range, the central Italy, to gain insights into the effects of this compensation practice on the genetic structure of its populations. Partridges from both a geographically isolated, long-time protected, wild population (Pianosa island, Tuscan Archipelago National Park) and two Tuscan farms (Bieri and Scarlino) were investigated. All the specimens were very similar in outward appearance, looking much like to A. rufa. Ninety-six sequences of both Cytochrome b and D-loop Control Region of mitochondrial DNA (mtDNA) were analysed to get evidence of ancestry at the population level, whereas, the Random Amplified Polymorphic DNA (RAPD) technique was employed to get fingerprinting at the individual level. Pianosa and Bieri populations showed both the A. rufa and Alectoris chukar mtDNA lineages, whereas the Scarlino one only the A. rufa-mtDNA line. However, a spread overall pattern of A. rufa × A. chukar hybridisation among specimens, whatever their mtDNA lineage could result to be, was disclosed by means of RAPD species-specific markers. This is the first genetically documented record of the A. rufa × A. chukar hybrids. The occurrence of the pure, native A. rufa genome in the easternmost part of the species’ geographical range may be guessed to be virtual.  相似文献

Recently, large areas of tropical peatland have been converted into agricultural fields. To be used for agricultural activities, peat soils need to be drained, limed and fertilized due to excess water, low nutrient content and high acidity. Water depth and amelioration have significant effects on greenhouse gas (GHG) production. Twenty-seven soil samples were collected from Jabiren, Central Kalimantan, Indonesia, in 2014 to examine the effect of water depth and amelioration on GHG emissions. Soil columns were formed in the peatland using polyvinyl chloride (PVC) pipe with a diameter of 21 cm and a length of 100 cm. The PVC pipe was inserted vertically into the soil to a depth of 100 cm and carefully pulled up with the soil inside after sealing the bottom. The treatments consisting of three static water depths (15, 35 and 55 cm from the soil surface) and three ameliorants (without ameliorant/control, biochar+compost and steel slag+compost) were arranged using a randomized block design with two factors and three replications. Fluxes of carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) from the soil columns were measured weekly. There was a linear relationship between water depth and CO2 emissions. No significant difference was observed in the CH4 emissions in response to water depth and amelioration. The ameliorations influenced the CO2 and N2O emissions from the peat soil. The application of biochar+compost enhanced the CO2 and N2O emissions but reduced the CH4 emission. Moreover, the application of steel slag+compost increased the emissions of all three gases. The highest CO2 and N2O emissions occurred in response to the biochar+compost treatment followed by the steel slag-compost treatment and without ameliorant. Soil pH, redox potential (Eh) and temperature influenced the CO2, CH4 and N2O fluxes. Experiments for monitoring water depth and amelioration should be developed using peat soil as well as peat soil–crop systems.  相似文献

Six triterpenoid saponins were isolated from the edible grain quinoa, which is seeds of Chenopodium quinoa (Chenopodiaceae). Following are their structures: phytolaccagenic acid 3-O-[alpha-L-arabinopyranosyl-(1' '-->3')-beta-D-glucuronopyranosyl]-28-O-beta-D-glucopyranoside (1); phytolaccagenic acid 3-O-[beta-D-glucopyranosyl-(1' '-->3')-alpha-L-arabinopyranosyl]-28-O-beta-D-glucopyranoside (2); phytolaccagenic acid 3-O-[beta-D-glucopyranosyl-(1' "-->3' ')-beta-D-xylopyranosyl-(1' '-->2')-beta-D-glucopyranosyl]-28-O-beta-D-glucopyranoside (3); phytolaccagenic acid 3-O-[beta-D-glucopyranosyl-(1' "-->2' ')-beta-D-glucopyranosyl-(1' '-->3')-alpha-L-arabinopyranosyl]-28-O-beta-D-glucopyranoside (4); oleanolic acid 3-O-[alpha-L-arabinopyranosyl-(1' '-->3')-beta-D-glucuronopyranosyl]-28-O-beta-D-glucopyranoside (5); and oleanolic acid 3-O-[beta-D-glucopyranosyl-(1' '-->3')-alpha-L-arabinopyranosyl]-28-O-beta-D-glucopyranoside (6). The oleanane-type saponins (5, 6) were isolated for the first time in this plant, two of the phytolaccagenane (1, 3) were new compounds and two (2, 4) were previously found in quinoa. The structures were characterized on the basis of hydrolysis and spectral evidence, including 1D- and 2-D NMR (HMQC and HMBC) and ESI-MS analyses.  相似文献
